Tadalaphyl is an efficient inversive selective inhibitor of specific phosphodiesterase of type 5 (PDE-5) of cyclic guanosine monophosphate (cGMP). Tadalaphyl improves erection, likewise the possibility of a successful sexual intercourse. The drug acts during 36 часов. The effect develops as quickly as in 16 minutes after the drug intake, in case of a sexual excitement.
Tadalaphyl does not provoke a firm change in frequency of heart beats. Tadalaphyl does not provoke changes in colour discrimination (blue/green), which is explained by its low affinity to PDE-6. Moreover, effect of tadalaphyl on vision, electroretinogram, tension on eye and the pupil dimension is not observed.
In the studies of Tadalaphyl use in men with a day’s dose of 10 mg. there was not revealed any drug’s significant influence on the quality and concentration of the sperm, agility and morphology of spermatozoids. Pharmacokinetics.
After intake by mouth Tadalaphyl absorbs quickly. Average maximum concentration (Сmax) in plasma is reached in about 2 hours after intake by mouth. Velocity and grade of the absorption of Tadalaphyl do not depend on meals intake that is why Tadalaphyl is not efficient if there is no sexual stimulation.
- Posology and Administration.
Cialis may be taken regardless of meals. The time of intake (morning or evening) does not give any clinically important effect on speed and grade of absorption.
Cialis pills should be taken orally.
The recommended dose of Cialis is 20 mg. Cialis should be taken before a supposed sexual activity. The drug should be taken as minimum 15 minutes before a supposed sexual activity.
Patients may try to start a sexual intercourse in any time during 36 hours after intake of the drug, in order to fix an optimum time of response to the drug intake.
Maximum recommended frequency of intake is once in 24 hours.
Elderly men do not need any special dosage. Recommendations on administration of the drug for middle-aged men are also recommendable for elderly patients.
- Counterindications.
Allergic reactions on any component of the drug,
determined high sensitivity to Tadalaphyl and any substance being a part of the drug;
in case of the use of the drug containing any organic nitrates;
use in people under 18 years old.
As there are no data of controllable clinic studies on safety and efficiency of Cialis for the following groups of patients: patients with severe kidney failure (clearance of creatinine is 730 ml/min), patients with severe liver failure, - such patients should take the drug with care.
- Should be Taken With Care If…
Sexual activity is potentially risky for patients with cardio-vascular diseases. That is why the treatment of erectile dysfunction, including the use of Cialis, should not be done in men with the heart illnesses where sexual activity is not recommended. One must consider a potential risk of aggravations in patients with cardio-vascular diseases: - myocardial infarction during the last 90 days; - unstable stenocardia or stenocardia emerging after a sexual intercourse; - heart failure of class 2 and higher in terms of NYHA, developed during the last 6 months; - uncontrollable disruptions of heart rhythm, hypotension (with AT <90/50 mm of merc. col.) or uncontrollable arterial hypertension; - insult underwent during the last 6 months.
- Interface with Other Medications.
Safety and efficiency of combination of Cialis with other kinds of the treatment of disruptions of erectile functions have not been studied yet. That is why the use of such combinations is not recommended.
It is counterindicated to administer Cialis together with the use of nitrates.
- Overdosing.
In a single-shot of Tadalaphyl with a dose up to 500 mg to healthy people and to patients with erectile dysfunction, or a multi-shot up 100 mg/a day - undesired effects were the same as in the use of lower doses. In a case of overdosing it is necessary to do a standard symptomatic treatment.
- Side effects.
The most frequently noticed undesired effects are headache and dyspepsia (11 and 7% cases respectively). Undesired phenomena linked with Tadalaphyl use usually were insignificant or medium in terms of manifestation rate, transitory and became less with continuation of use of the drug. Other usual undesired effects are aching back, myalgia, stuffed nose and "flushes" of blood to the face. Seldom – hydrops of eyelids, aching eyes, hyperemia of conjunctiva and dizziness.
- Pregnancy and Lactation.
Cialis is not indicated for use in women
- Driving.
There are no special indications.
- The expiry date is mentioned on each blister. It is different for different batches. The shelf life is 2 years from the date of manufacture and would differ from batch to batch depending on when they were manufactured.
